Do I need to be connected to wifi to update all the books in my library onto my nook? I can hook it up to a computer with an internet connection, but that doesn't seem to be working...

If you want the books to be in your My B&N library on the Nook then you need to be connected to WiFi. Connecting to a computer doesn't connect the Nook to the internet. When you connect the Nook to the computer it is just like connecting any type of external drive, such as a flash drive.
You can download the books to your computer and sideload to the Nook if you don't have access to WiFi, there's instructions for sideloading on the Nook support page.

Yes you need to have WIFI or 3G to update the books on the Nook no connect to the PC. Once you know you have WIFI connection in the B&N library click on check for new content and that should update all your books but if you have a lot, it may take a couple times. On my Nook color it shows all 380 books but has a DL symbol beside them.
