The Nook Family supports MP4 video so check to ensure that your clips are in fact in MP4 format. If you do not have a video converter on your computer a very capable, easy to use and optimized for Nook converter is DVD Catalyst which you can get on-line for about $9 from Tools4Movies.com. I like this converter because if your choose the Nook Tablet or Color profile it does every setting to provide an excellent quality video.

If the movie clips are DRM protected, you can only play them on Apple devices.

I have now noticed that if I open the video on nook when it shows that it finished downloading to the SD card I can watch the video. When I try to watch the same video from My Media it shows it with a ? on it and when I click on it it says that it is not supported. The videos I am talking about is short ones of family taken with an Iphone. When a Blackberry was used for family videos I had no trouble.They both are MP4...

The "My Media" app only likes to play videos that are located in the Videos folder. If they are supported video files, you should be able to play them using My Library if they are located somewhere else.

I go to library, my files, my memory card and then click on the video. I get" The nook cannot read this file. It may be an unsupported format"
Like I said I can see the video fine when it is first downloaded to my memory card and i click on the symbol that it has finished downloading...a bunch of arrows on the left side of the memory card. After that I have tried opening them from my library and won't open.

As GeertM mentioned, videos you buy/rent from iTunes are mostly DRM-protected. I guess I misread your last reply:

From your reply, it seemed that the video file would actually play on your NOOK, but you probably meant that when you copy the file over to your NOOK and tapped the video icon on your computer it would play on your computer, which is to be expected, since the video file is activated on your computer, and the NOOK, when connected with the USB cable, is just seen as a storage device.
The thing is, iTunes movies are copy protected. Apple doesn't want you to use those movies on devices that Apple didn't create, so they implemented something that prevents you from using them on anything that doesn't have an Apple logo.

I just downloaded the video from my email....msn... through Dolphin...the web browser...right on my nook tablet. No computer used...

Try reconverting the MP4s into MP4s and see if that helps.
