It's not a stupid question.
The best option would be:
1) Charge your Nook to at least 70%.
2) Turn on WiFi.
3) Press the power button to put it to sleep.
Then the Nook will automatically check for updates and will update.

I'm not sure about whether in manual updates it's OK to go from 1.4.1 to 1.4.3.
It should be.
Finally, there's not much added in 1.4.3. I would recommend letting the automatic update happen (put your device to sleep with battery charged) at its own time.
